Consequently, the rise of internal sodium causes via the sodium-calcium exchanger an elevated calcium ion level leading to the hypertension producing vasoconstriction. Es wird geschätzt, dass fast 10% der Weltbevölkerung an einer solchen Infektion leiden. Gerade bei den chronischen Hepatitiserkrankungen werden oft kaum Antikörper gebildet, um die Viren zu bekämpfen. Doch die Schwere der Erkrankung ist in vielen Fällen nicht allein dem Virus selbst und einer mangelhaften Immunantwort zuzuschreiben. Oft kommen fehlgeleitete Abwehrreaktionen hinzu, bei denen sich Antikörper gegen das eigene Gewebe wenden. Woran dies liegt, erforscht Philipp Alexander Lang. Hierzu nutzt er zwei verschiedene Infektionsmodelle. Einmal Infektionen, die durch ein zellschädigendes Virus erfolgten, und solche, an denen ein Virus Schuld ist, das keine Zellschäden verursacht. Im Vergleich beider Infektionsmodelle will Lang herausfinden, wie die Bildung solcher Antikörper begünstigt wird, die tatsächlich das Virus wirksam attackieren und nicht den eigenen Körper. Infectious diseases, such as HIV, viral hepatitis, malaria, and tuberculosis are amongst the most prevalent causes of death. The human body has developed a powerful immune system, which efficiently eliminates pathogens. However, the rigorous battle may cause collateral damage to our own cells, which even can exceed the damage caused by the pathogen itself. Moreover, our immune system may erroneously consider our own cells and structures as foreign, by mistake attack those and thus cause autoimmune disease, such as type I diabetes. The research of Dr. Lang aims to unravel the causes for errors of the immune system. In the past, he has discovered why viral disease could trigger type I diabetes. He now explores why some 20% of patients with hepatitis do not eliminate the virus but may end up with complete destruction of this essential organ. His most recent research discloses that the course of hepatitits critically depends not only on the immune system, but also on specific genes of liver cells. Dr. Lang hopes that understanding the causes of error will allow restraining the immune system and thus minimize collateral damage and self-destruction. Das häufig auf dem Boden der Zirrhose entstehende Leberzellkarzinom ist der fünfthäufigste Krebs weltweit. Die Leber als größtes abdominelles Organ des Körpers erfüllt nicht nur im Stoffwechsel, sondern auch in der Immunabwehr vielfältige Aufgaben, die bisher unvollständig aufgeklärt sind. Frau Dr. med. (HU) Dr. rer. nat. Lukacs-Kornek hat während ihrer Postdoktorandenzeit in der Arbeitsgruppe von Shannon Turley an der Harvard Medical School die Rolle der fibroblastischen retikulären Zellen (Stromazellen) im lymphatischen Gewebe aufgeklärt. An der Universität des Saarlandes möchte die neu ernannte Professorin den Aufbau und die Funktion des tertiären lymphatischen Gewebes (TLT) der Leber untersuchen und die Bedeutung der Stromazellen für die Entstehung chronischer autoimmun-vermittelter und biliärer Lebererkrankungen klären. Dieses Vorhaben ist klinisch von großer Relevanz, da bisher für viele chronische Lebererkrankungen - wie die als Modellerkrankung ausgewählte primär-sklerosierende Cholangitis (PSC) -außer der Transplantation keine effektiven Behandlung zur Verfügung steht. Die Rolle der Antigenpräsentation durch Stromazellen soll nicht nur in präklinischen Krankheitsmodellen, sondern auch bei der Krebsentstehung in der chronisch entzündeten Leber untersucht werden. Auf diese Weise erweitert sich nicht nur unser Verständnis der immunologischen Aufgaben der Leber, sondern es eröffnen sich neue Möglichkeiten, frühzeitig die Entwicklung und das Fortschreiten chronischer Lebererkrankungen zu verhindern. As evident by a prominent infiltration with immune cells, it is now widely accepted that atherosclerosis is a chronic inflammatory disease with a pivotal role for the innate and adaptive immune system. The co-stimulatory molecules CD40 and CD40L responsible for expansion and differentiation of naive T-lymphocytes into effecor-cells have been identified as key players in atherosclerotic plaque progression. Since the clinical complications of atherosclerosis mostly result from plaque rupture, induction of plaque stability will reduce the morbidity and mortality of atherosclerosis. Professor Lutgens hypothesizes that targeting recently identified atherosclerosis-specific components of the CD40-CD40L pathway, e.g. CD40-TRAF6, will be a valuable therapeutic strategy to induce stable plaques without immune-suppressive side-effects. The downstream pathways of CD40 and their therapeutic suitability will be elucidated in atherosclerotic mice, in which components of the CD40-pathway have been modified. Epidemiologie und Public Health

Hängen der Klimawandel und die Ausbreitung von Krankheiten zusammen – und wenn ja, wie? In dieser zum Teil kontrovers geführten Diskussion stellt Joacim Rocklöv wichtige Daten bereit und liefert Analysen für die Modellierung möglicher zukünftiger Szenarien – die essenzielle Grundlage für politische Entscheidungen.
Die Untersuchung des Klimawandels und seiner Auswirkungen gehören zu den großen Herausforderungen unserer Zeit. Joacim Rocklöv arbeitet stark interdisziplinär im Bereich Klimawandel, Infektionsepidemiologie und Modellierung. Gesundheitliche Auswirkungen des Klimas sollen in der Zukunft besser abgeschätzt werden können, ein Ziel sind Vorhersagemodelle für Frühwarnsysteme und die bessere Kontrolle von Infektionskrankheiten. Rocklövs Ergebnisse basieren auf einer innovativen Kombination von Daten verschiedener Fachgebiete aus der ganze Welt mit Methoden von KI, Data Mining und Statistik.

In Heidelberg soll jetzt das erste KI-Labor in Deutschland zu Fragen von globalen Infektionskrankheiten und Klimaveränderungen entstehen. Rocklövs Humboldt-Professur wird an der Schnittstelle von zwei Instituten installiert: dem Heidelberg Institute of Global Health (HIGH) der Medizinischen Fakultät Heidelberg, das von Humboldt-Professor Till Bärnighausen geleitet wird, und dem Interdisziplinären Zentrum für Wissenschaftliches Rechnen (IWR). Damit will die Universität Heidelberg ihre bestehende nationale Spitzenposition im Bereich der methodengestützten Global Health-Forschung weiter ausbauen und auch in die international Spitze vorrücken. Außerdem soll eine bisher fehlende Verknüpfung zur klinischen Infektionsforschung in Heidelberg möglich werden.

Joacim Rocklöv wurde für die Humboldt-Professur ausgewählt und ist derzeit in Berufungsverhandlungen mit der deutschen Universität, die ihn für den Preis nominiert hat. Werden diese erfolgreich abgeschlossen, wird der Preis 2022 verliehen.

Zur Person

Prof. Dr. Joacim Rocklöv wurde in Schweden geboren und ist seit 2018 Professor am Department für Public Health und Klinische Medizin der Umeå Universität in Umeå, Schweden, wo er zuvor bereits Senior Lecturer und dann Associate Professor war. 2010 wurde er promoviert. 2012 erhielt er den Young Researcher Award der Umeå Universität, 2019 wurde ihm der Prince Albert II of Monaco - Institut Pasteur-Preis verliehen für seine Forschung zu klimabedingten Arbovirus-Epidemien. Er ist Mitherausgeber von Fachzeitschriften, berät internationale Gremien in den Bereichen Epidemiologie und Klimawechsel und hat an einem Tropenkrankheiten-Frühwarnsystem der Weltgesundheitsorganisation mitgearbeitet.
