His research activities reach into many areas of the Neurosciences ranging from the molecular mechanisms of neuronal fate determination to the circuitry underlying behaviour. He identified a group of genes that participate in the decision whether cells in the early mouse embryo become neurons or epithelium. In 1989 he proposed the existence of self-renewing stem cells in the nervous system and a few years later his group achieved the first isolation of such cells. Moreover, by transplantation experiments he demonstrated that the isolated cells can generate both neurons and glia. Prof. Anderson's group also identified some of the growth factors promoting this differentiation. In fact, they provided the first clear demonstration, in any system, that growth factors can instruct the fate of multipotent stem cells. In 1998, Prof. Anderson and colleagues reported the surprising and remarkable discovery that the endothelial cells of developing arteries and veins exhibit differences in gene expression that can be detected already in the very earliest stages of vascular development, before the heart starts to beat. These findings led Prof. Anderson to the now well established hypothesis that reciprocal interactions between arteries and veins are essential for the development of the vascular system. Prof. Anderson has combined his interests in the development of the nervous and vascular systems to address a long-standing question about the relationship between nerve- and blood-vessel branching. He revealed that peripheral sensory nerves in the skin are aligned with arteries but not veins, and that this alignment even extends to the finest-diameter blood vessels. More recently, Prof. Anderson has developed an active interest in behavioural neuroscience. He has started to investigate stress- and possibly fear-related innate behaviour and its neuronal substrate in flies. Collaborations in this field have guided his choice of the host laboratory in Germany, the group of behavioural neurogenetics around Martin Heisenberg at the Biocenter of the University of Würzburg. There, he develops behavioural assays for studying the valuation process by which flies decide between the putative benefits of different behavioural options. He intends to eventually relate this process to genes, modulator molecules and neuronal circuitry. At the same time, in his home laboratory at the California Institute of Technology, he is gearing up to approach by similar neurogenetic methods the vastly more complex behaviour and brain of the mouse. Nicht die Gene selbst verursachen folglich die Unterschiede, sondern ihre Steuerung über sogenannte epigenetische Markierungen im Erbgut. Diese steuern die Aktivität von Genen und spielen so für die Ausbildung unterschiedlicher Verhaltensweisen und Äußerlichkeiten eine zentrale Rolle. Ameisen sind also für die Erforschung der epigenetischen Steuerung im Gehirn ideale Untersuchungsobjekte. Roberto Bonasio hat das Erbgut der Ameisenart Harpegnathos saltator mit einer sehr hohen Genauigkeit entschlüsselt. Die Art hat sich seitdem zu einem Modellorganismus entwickelt, an dem Forschende die Auswirkungen epigenetischer Veränderungen untersuchen können. Harpegnathos saltator kommt in Indien vor und wird zwei bis drei Zentimeter lang. Die Tiere leben in kleinen Kolonien mit einer Königin, die sich zwar äußerlich nicht sehr von den anderen Mitgliedern der Kolonie unterscheidet, dafür aber im Verhalten. Ausgewachsene Harpegnathos-Arbeiterinnen haben die unter Ameisen seltene Fähigkeit, sich in sogenannte Pseudo-Königinnen umzuwandeln. Sie verhalten sich dann wie echte Königinnen, legen Eier und leben länger. Bonasio und sein Team können diese Verwandlung im Labor künstlich anstoßen und so die veränderte Genaktivität der Tiere analysieren. Auf diese Weise können die Forschenden die epigenetischen Markierungen, Transkriptionsfaktoren – Proteine, die die Aktivität von Genen steuern – und neuronalen Botenstoffe untersuchen, die für diesen Kastenwechsel verantwortlich sind. Sie haben hunderte von Genen identifiziert, die bei der Umwandlung von Arbeiterinnen in Pseudo-Königinnen im Gehirn der Tiere ihre Aktivität ändern. Besonders das Gen für Corazonin, ein aus wenigen Aminosäuren bestehendes Peptid, ist dabei besonders stark im Gehirn der Arbeiterinnen aktiv. Mit einer Reihe ausgeklügelter Experimente haben Bonasio und sein Team damit nachgewiesen, dass es für verschiedene Verhaltensweisen der Arbeiterinnen verantwortlich ist, zum Beispiel die Jagd und Nahrungsbeschaffung für die Königin. Aufgewachsen in Sesto Calende am Lago Maggiore studierte Bonasio zunächst Biotechnologie in Mailand. Nach Stationen in Harvard und an der Universität New York, leitet er seit 2014 sein eigenes Labor an der Perelman School of Medicine der Universität Pennsylvania. Mit der Auszeichnung verbunden ist auch ein Forschungsprojekt, für das er am Universitätsklinikum Freiburg (UKF) ein eigenes Labor aufbauen wird. Gemeinsam mit dortigen Wissenschaftlern um den Neuropathologen Marco Prinz (UKF) und den Epigenetiker Marc Timmers (UKF/DKTK) will Bonasio die Umstrukturierung des Gehirns bei Individuen genauer untersuchen, die zwischen Arbeiterinnen- und Königinnenstatus wechseln. Es ist genau diese Formbarkeit des Gehirns, die den Ansatz so vielversprechend macht für Anwendungen beim Menschen. Milliarden von Nervenzellen und Billionen von Verknüpfungen, die Synapsen, erlauben die Kontrolle vitaler Funktionen von der Atmung bis zur Ausübung vielschichtiger mentaler Aufgaben. Unablässig muss sich das Gehirn auf Veränderungen in der Umwelt einstellen. Ein Schlüssel hierfür ist die synaptische Plastizität, die Fähigkeit der Synapsen, sich und die Stärke der von ihnen übertragenen Signale anzupassen. Steigt die Signalstärke jedoch zu sehr an, kann es zu Schädigungen oder epileptischen Anfällen kommen. Camin Dean untersucht die Mechanismen, die die Synapsen in der Balance halten, so dass das Gehirn funktionieren kann. Sie hat herausgefunden, welches Protein die Stärke von Synapsen auf einen sinnvollen Bereich einstellt, indem es die Freisetzung eines Neurotrophins kontrolliert, eines Signalstoffes, der nicht nur die synaptische Plastizität, sondern auch die Bildung neuer Synapsen beeinflusst. Dean will die zugrunde liegenden Mechanismen studieren und so dazu beitragen, Erkrankungen wie die Alzheimer-Krankheit, Morbus Parkinson und Epilepsie besser behandeln zu können.
